This research project aims to investigate the interactions of phytoestrogens with oestrogen receptors in breast, uterine and bone cells.
Study Duration : March 2002 to February 2004
Contractor : University of Reading
In humans, compounds with oestrogenic properties act via two receptors (ER alpha and ER beta). The tissue distributions of these receptors are different. Phytoestrogens are known to bind more strongly to ER beta than ER alpha and so they may have different effects on different tissues. This project aims to examine the action of phytoestrogens at ER beta in a number of different human cell lines.
The effects of ER beta overexpression on the regulation of gene expression by phytoestrogens in breast, uterine and bone cells will be examined.
Over expression of ER beta reduced induction of CAT (a stably integrated reporter gene) activity by estrogen, daidzein, equol, coumestrol and 8-prenylnaringenin, but enhanced gene expression with high concentrations of genistein and resveratrol. Further work to enhance the overexpression of ER beta would be needed to establish the significance of these findings.
The final report is available from the Agency's Information Centre.
